Overview

Genotoxic impurities are substances that are capable of directly or indirectly damaging cellular DNA, thereby causing DNA mutations and associated carcinogenicity risks. The damaging effects of such impurities on DNA include chromosome breakage, insertion and modification of covalent bonds during DNA recombination and replication, among others.

Classification of Genotoxic Impurities

Class 01

Known mutagenic carcinogens

Class 02

Known mutagens with unknown carcinogenic potential (bacterial mutagenicity positive,a no rodent carcinogenicity data)

Class 03

Alerting structure, unrelated to the structure of the drug substance; no mutagenicity data

Class 04

Alerting structure, same alert in drug substance or compounds related to the drug substance (e.g., process intermediates) which have been tested and are non-mutagenic

Class 05

No structural alerts, or alerting structure with sufficient data to demonstrate lack of mutagenicity or carcinogenicity
